Launched in 1977, Voyager 1 is now poised to become the first craft to enter interstellar space.

NASA's Voyager 1 spacecraft has encountered a new environment more than 11 billion miles from Earth, suggesting that the venerable probe is on the cusp of leaving the solar system.

The Voyager 1 probe has entered a region of space with a markedly higher flow of charged particles from beyond our solar system, researchers said. Mission scientists suspect this increased flow indicates that the spacecraft ? currently 11.1 billion miles (17.8 billion kilometers) from its home planet ? may be poised to cross the boundary into interstellar space.

"The laws of physics say that someday Voyager will become the first human-made object to enter interstellar space, but we still do not know exactly when that someday will be," said Ed Stone, Voyager project scientist at the California Institute of Technology in Pasadena, in a statement.

"The latest data indicate that we are clearly in a new region where things are changing more quickly," Stone added. "It is very exciting. We are approaching the solar system's frontier." [Photos From NASA's Voyager 1 and 2 Probes]

Far-flung spacecraft

Voyager 1 and its twin, Voyager 2, launched in 1977, tasked chiefly with studying Saturn, Jupiter and the gas giants' moons. The two spacecraft made many interesting discoveries about these far-flung bodies, and then they just kept going, checking out Uranus and Neptune on their way toward interstellar space.

They're not quite out of the solar system yet, however. Both are still within a huge bubble called the heliosphere, which is made of solar plasma and solar magnetic fields. This gigantic structure is about three times wider than the orbit of Pluto, researchers have said.

Paramedics call for new union reps

Ambulance workers say the Health Services Union is a problem and they don't want to be forced to join.

Carrying placards with the slogan 'Freedom of Choice, Give us a Voice', more than 100 off-duty ambulance workers have called to be permitted to seek industry-specific industrial representation.

Currently paramedics can call on only the HSU as their official union representatives.

However, as the union has become engulfed in scandal in recent years, many ambulance workers have ended their membership and joined the fledgling Emergency Medical Service Protection Association (EMSPA).

But EMPSA spokesman Wayne Flint says that because the union is not industrially registered paramedics have been left without a voice.

'The HSU is a problem, we don't want to be a part of it,' Mr Flint told reporters on Tuesday.

'We haven't had good representation from the HSU in our view, and paramedics as the most trusted profession deserve a voice.'

A bill that would give EMSPA industrial registration is stuck in the NSW Upper House, where the coalition, Labor and minor parties are unable to agree on proposed amendments.

NSW Health Minister Jillian Skinner has called for the amendments to be passed before the start of parliament's winter recess this week.

'The whole point of our legislation is to give freedom of choice to people about what union they should have represent them,' Ms Skinner told the rally.

A report commissioned by the HSU and conducted by Ian Temby QC on April 30 found the HSU East branch awarded $17 million of dodgy tenders and had no credit-card policies.

Eastern branches of the HSU were last week placed into administration and its officers suspended by a Federal Court judge.

Wisconsin Recount Ordered In State Senate Race Between John Lehman, Van Wanggaard

Wisconsin's board of elections has approved a request from state Sen. Van Wanggaard (R-Racine), officially ordering a recount in a recall election that some had thought had been settled in Wisconsin's 21st district.

The Associated Press reports that the state's Government Accountability Board issued the order on Monday afternoon, asking county officials to begin the process at 9 a.m. local time on Wednesday.

Democrats had earlier called on Wanggaard to concede to his recall challenger, Democrat John Lehman, who had already declared victory. The Racine County clerk had also certified the win, but Wanggaard's campaign had expressed concerns about the accuracy of the close vote, which showed the two candidates separated by about 834 votes, or 1.2 percent of around 72,000 votes cast. Wanggaard requested the recount on Friday.

The race holds statewide and national implications because it will decide which party controls the state Senate, which is split at 16 senators for each party, pending the results of this race. Furthermore, Lehman's apparent triumph was held up as a bright spot by Democrats who experienced a massive disappointment in the broader recall effort when they failed in their effort to recall Wisconsin Gov. Scott Walker (R) and other Republicans.

Racine County will have until July 2 to complete the recount process.

Antitoxin strategy may help target other pathogens

Researchers have unveiled a novel strategy for neutralizing unwanted molecules and clearing them from the body.

The strategy employs chains of binding agents, like "beads on a string", which target two sites on one or more pathogenic molecules to neutralize their activity and promote their clearance by the body's immune system. The low-cost, easy-to-replicate tool has demonstrated applications against several different toxins, from those found in contaminated food to those used in bioterrorism, and may also prove effective in targeting other types of pathogens.

The research team, based at Tufts University's Cummings School of Veterinary Medicine, demonstrated the method's efficacy in preventing the symptoms of botulism, a rare but deadly disease caused by Clostridium botulinum neurotoxin (BoNT), considered one of the most dangerous bioterror threat agents. The findings were presented earlier this year in PLoS ONE.

"Currently, antitoxins are difficult to produce and have a short shelf life, making them very expensive. This new approach provides a low-cost way to develop highly effective antitoxins," said senior author Charles B. Shoemaker, PhD, professor of biomedical sciences at Tufts University's Cummings School of Veterinary Medicine.

"This method has the potential to target a number of pathogens ? not only toxins such as BoNT, but viruses or inflammatory cytokines. It is an important platform through which to address other significant diseases," says co-author Saul Tzipori, BVSc., DSc, PhD, professor of biomedical sciences and director of the Division of Infectious Diseases at the Cummings School.

Shoemaker and team had earlier found that pools of small 'tagged' binding agents were highly effective in targeting toxins, neutralizing their function, and flagging them for removal via the body's immune system in the presence of an anti-tag monoclonal antibody.

In the newly published in vivo study, the researchers have advanced this approach by linking two BoNT-binding agents together and including two copies of the tag. The binding agents are small, stable proteins derived genetically from unusual antibodies produced by toxin-immune alpacas. The resulting molecule, called a 'double-tagged heterodimer,' binds to two separate sites on the toxin. Binding of this single heterodimeric agent much more effectively neutralizes the toxin than the unlinked monomer binding agents used in the prior research. In addition, attaching two tags to each of the two linked agents leads to toxin decoration by up to four anti-tag monoclonal antibodies, which promotes rapid toxin clearance from the blood, the researchers found (see figure).

The double-tagged heterodimer antitoxin agent strategy was shown to be efficacious against two types of BoNT in the PLoS ONE report. The antitoxin agents were administered at the time of exposure, or shortly after. Treated mice did not show any symptoms of botulism ? including the lethal paralysis which characterizes the disease, even when exposed to high toxin doses. Thus, the benefits of complex antitoxins were equaled or bettered by administration of two easy-to-produce agents; a heterodimer binding agent and an anti-tag monoclonal antibody.

According to Shoemaker, a major advantage of this approach is that, unlike treatments that only neutralize toxins, this treatment both neutralizes toxins and ensures their rapid clearance from the body. "Agents that only neutralize their pathogenic target will eventually dissociate which will allow the pathogen to continue doing damage if it is not eliminated," he said.

The group has now successfully taken the research further by building longer strings of binding agents that target multiple toxins with a single molecule?for example, the two types of Shiga toxins that are produced by some E. coli found in contaminated foods or the two toxins produced by hospital-acquired C. difficile infections.

Facebook settles sponsored stories suit for $10M

Facebook is paying $10 million to settle a lawsuit over ads that it called sponsored stories.

Facebook's service allowed companies to pay to retransmit users' activities to their friends' pages. If a user clicked the "like" button for a brand, the click might show up as a "sponsored story" on friends' pages. The ads started in early 2011.

Some Facebook users sued, saying that Facebook used their images for commercial activity. Facebook had argued that the plaintiffs did not show they were hurt by the practice.

Court documents from late last week say a settlement has been reached. Facebook attorney Michael Rhodes confirms Sunday that it's for $10 million, which will be given to charity. A judge still needs to approve the settlement.

Lariats: How RNA splicing decisions are made

ScienceDaily (June 17, 2012) ? Lariats are discarded byproducts of RNA splicing, the process by which genetic instructions for making proteins are assembled. A new study has found hundreds more lariats than ever before, yielding new information about how splicing occurs and how it can lead to disease.Tiny, transient loops of genetic material, detected and studied by the hundreds for the first time at Brown University, are providing new insights into how the body transcribes DNA and splices (or missplices) those transcripts into the instructions needed for making proteins.

The lasso-shaped genetic snippets -- they are called lariats -- that the Brown team reports studying in the June 17 edition of Nature Structural & Molecular Biology are byproducts of gene transcription. Until now scientists had found fewer than 100 lariats, mostly by poring over very small selections of introns, which are sections of genetic code that do not directly code for proteins, but contain important signals that direct the way protein-coding regions are assembled. In the new study, Brown biologists report that they found more than 800 lariats in a publicly available set of billions of RNA reads derived from human tissues.

"We used modern genomic methods, deep sequencers, to detect these rare intermediates of splicing," said William Fairbrother, associate professor of biology and senior author of the study. "It's the first ever report of these things being discovered at a genome scale in living cells, and it tells us a lot about this step of gene processing."

That specific step is known as RNA splicing. Like film editors splicing together movie scenes, enzymes cut away the introns to assemble exons that instruct a cell's ribosome to make proteins. The body often has a choice of ways and places to make those cuts. Most of what is known about splicing has come from studying these spliced instructions, said Allison Taggart, a graduate student who is lead author of the study. What's been missing is the data hidden in the lariats, which fall apart shortly after being spliced out, but turn out to predict the body's splicing choices.

Modeling splicing

The key information uncovered in the study, Taggart said, is the location of so-called "branchpoints" on the lariats. Physically, the branchpoint is where the lariat closes on itself to form a loop during the first step of splicing, but its position and proximity to possible splice sites, the researchers learned, reliably relate to where splicing will occur.

After studying the sites of these branchpoints and their relationship to splice sites, the researchers created an algorithmic model that could predict splice sites 95.6 percent of the time. The value of the model is not in identifying splice sites -- those are already well known, Fairbrother said. Instead, the model's accuracy shows that, with the new data from the lariats, scientists have gained a more general understanding of how the body chooses among alternative splicing sites.

"What it does tell us is sets of rules defining the relationship between branchpoints and the chosen splice sites, which gives clues about how the splicing machinery makes decisions," Taggart said. "Certain branchpoint locations can enforce specific splicing isoforms."

Connections to disease

In addition to ferreting out the mechanisms of alternative splicing, the team also studied the connection between branchpoints and disease. They looked through the Human Gene Mutation Database for disease-causing mutations found in introns and compared their newly found branchpoint sequences to those mutations. They found that many relate specifically to branchpoints.

"We saw a sequence motif that looked exactly like a branchpoint sequence motif," she said. "What this tells us is that these mutations are forming at branchpoints and are leading to disease, presumably through causing aberrant splicing by interfering with lariat formation."

In other words, Fairbrother said, it could well be that a consequence of mutations in branchpoints could be disease.

In addition to Taggart and Fairbrother, other authors include Alec DeSimone, Janice Shih, and Madeleine Filloux.

The National Science Foundation and Brown University funded the research, which was performed in part on the OSCAR supercomputing cluster at the University's Center for Computation and Visualization.
